Figure 5

Enhanced inhibition of forskolin-stimulated adenylate cyclase by lysophosphatidic acid following conditional induction of the Gα i2 G184S mutation in adult cardiac fibroblasts. Concentration-response curves were generated from adult cardiac fibroblasts isolated from three different mice of each genotype after treatment with tamoxifen (50 mg/kg i.p. on five consecutive days). Data from 5 independent determinations for both Gαi2 fl-G184S/fl-G184S and Cre-ERT2/Gαi2 *G184S/*G184S are shown as mean ± SE. Data are normalized to cAMP production in the presence of 10-13 M lysophosphatidic acid. Two way ANOVA revealed a significant effect for genotype [F = 5.58 (1,8) p < 0.05] and a significant effect for lysophosphatidic acid concentration [F = 12.09 (8, 64) p < 0.0001]. * indicates a significant difference (p < 0.05 compared to Gαi2 fl-G184S/fl-G184S cells treated with 10-6M LPA) using Bonferroni’s post test.
